A sensitive method for the determination of vinblastine in rat blood plasma and tumour tissue using flow cytofluorometry

A method is described which allows the determination of low concentrations of vinblastine in blood plasma and tumours in experimental animals. This method is based on the biological activity of the compound and measures the compound itself and its biologically active metabolites. It employs the measurement of cell kinetic changes in cultured cells induced by tissue samples containing vinblastine. Analysis of these changes is carried out with a flow cytofluorometer. The minimum amount of vinblastine which can be detected is about 0.5 x 10-8 g/ml. The method is applied to determine vinblastine pharmacokinetics in WAG/Rij rats with a transplantable R-l rhabdomyosarcoma. Comparison of vinblastine pharmacokinetics with cell kinetic effects in tumours shows that the correlation is complex.
