Synthetic opioids are a group of highly potent compounds with analgesic and sedative properties, mediated by the ยต-opioid receptor (MOR). MOR activation is also associated with potentially life-threatening respiratory depression and muscle rigidity. In addition to the well-known synthetic opioid fentanyl, many structural analogues have been described. Due to their high potency and availability, synthetic opioids are considered a major (public) health concern. To understand the risks, it is essential to not only evaluate the potency of fentanyl and analogues, but also their intermediates, by-products and breakdown products. In this study, we selected, developed, and validated a luminescence-based assay for the screening of synthetic opioids and evaluated various in-house synthesized synthetic opioids and precursors.
