Preparation and characterization of Kupffer cells from rat and mouse liver

Enzymatic procedures for the preparation of nonparenchymal cells from rat and mouse liver are described and the results are compared. Optimal results with respect to yield and recovery of Kupffer cells were obtained after pronase treatment of the liver. For specific studies on membrane properties of Kupffer cells, a nonparenchymal cell preparation method based on collagenase treatment of the liver may be useful. Kupffer cells in nonparenchymal cell suspensions can be further purified by attachment during maintenance culture or by centrifugal elutriation. The latter method resulted in highly purified, viable Kupffer cell suspensions. Kupffer cells purified from rat liver were characterized by their ultrastructure and lysosomal enzyme content. It is concluded that the purpose for which the cells will be used determines the choice of the preparation procedure.
