A live attenuated malaria vaccine: Science or fiction?

In this article practical aspects of malaria vaccine development and application are discussed. The impact of polymorphism of the malaria parasite on vaccine development demands a re-evaluation of the feasibility of application of recombinant vaccines containing one or a limited number of parasite antigens. Some hypothetical possibilities for the generation of attenuated parasite lines are discussed and arguments in favour of this strategy are compared with the limitations. At the same time the scientific aspects of using gametocyte producing parasites for development of a self-spreading vaccine are examined. Chemicals/CAS: Protozoan Vaccines; Vaccines, Attenuated.
