Elastolipoproteinase: an eserine-resistant esterase? further purification and characterization of this pancreatic enzyme

1. 1. A purer elastolipoproteinase preparation has been prepared in order to get more information for the suggestion put forward in an earlier paper that this enzyme and the postheparin lipoprotein lipase are closely related to each other. 2. 2. The esterolytic activity of elastolipoproteinase and two other fractions which could be isolated from a pancreatic extract by column chromatography was studied on Tween 20 and normal serum. Several compounds were added to the reaction mixture and their inhibitory or activation activity measured. 3. 3. None of the three pancreatic enzyme fractions showed the activity of lipase, phospholipase A or C, ali-esterases, cholesterol esterase or cholesterol synthetase. 4. 4. From the results of these experiments it is concluded that our previous suggestion was wrong. Not elastolipoproteinase but the fraction eluted from the column with phosphate buffer pH 6.6 + 0.17 M NaCl shows the characteristics of the clearing factor lipase. 5. 5. Elastolipoproteinase behaves more like an eserine-resistant esterase and may act in this way on the lipomucoprotein moiety present in elastic tissue as a cross-linking agent. © 1970.
