New aspects of blood collection and handling procedures for the assessment of t-PA and PAI-1 variables after the introduction of acid blood collection

Blood collection and handling procedures for t-PA activity have thusfar been rather strict and have restrained the use in clinical practice. We evaluated simplifications in the procedures for specifically the use of Stabilyte® vacutainers with respect to time before centrifugation of blood, storage of plasma, times of inversion of the collection tube and the working temperature. For t-PA activity it was documented that blood storage up till 2 hours and plasma storage up till 3 hours, both at room temperature, gave no differences in results compared to the previous strict procedure at 4°C. It appeared also possible to measure reliably t-PA antigen and PAI-1 antigen in the Stabilyte samples, however, for PAI-1 antigen limited stability of platelets in blood require to restrict the blood storage time at room temperature to 30 minutes or to cool the blood to 4°C.
